Kinetics of calcium transport by fragmented sarcoplasmic reticulum.

چکیده
Calcium transport by sarcotubular vesicles (fragmented sarcoplasmic reticulum) from skeletal muscle of rats, rabbits, and humans follows Michaelis-Menten kinetics when studied in systems containing oxalate and low ratios of calcium to vesicle protein. Under these conditions different preparations of vesicles are characterized by their calcium concentrating ability, i.e. the constant level to which they lower the calcium concentration of the medium. K,,, and V,,, values for calcium transport by rat and human vesicles are now established, and the rates at which calcium leaks from vesicles at equilibrium are detied with the use of two different methods of studying exchange at equilibrium. Chlorpromazine, quinine, and quinidine are competitive inhibitors of calcium transport. Reduction of calcium to Michaelis-Menten kinetics should facilitate comparison of the calcium transport system in vesicles from various types of muscle.

Regulatory Mechanisms of the Calcium Transport System of Fragmented Rabbit Sarcoplasmic Reticulum
The outflux of calcium from vesicles of sarcoplasmic reticulum is controlled by the concentration of ionized calcium in the medium. In a calcium-free medium calcium outflux is at a minimum provided ATP and Mg are present. If they are removed calcium outflux becomes rapid and has a rate constant similar to that of outflux during steady-state (40)Ca-(45)Ca exchange with saturating calcium concent...
متن کاملEffect of physical training on calcium transport by rat cardiac sarcoplasmic reticulum.
Cardiac microsomes enriched in fragmented sarcoplasmic reticulum (SR) were isolated from hearts of physically trained rats and were compared to those from sedentary rats. Preparations from conditioned rats were found to transport calcium to a greater extent than those from sedentary rats both in the absence and in the presence of 1 mM oxalate. Higher oxalate concentrations abolished the differe...
متن کاملFormation and decay of the vanadate complex of the sarcoplasmic reticulum calcium transport protein.
The calcium free sarcoplasmic reticulum calcium transport ATPase incorporates in the presence of magnesium ions approx. 8 nmol monovanadate per mg protein, indicating the formation of a complex containing one vanadate residue per enzyme molecule. On ligand-removal or dilution, the saturated enzyme complex displays biphasic decay kinetics, while the unsaturated complex slowly dissociates monopha...

Fragmented sarcoplasmic reticulum (FSR) from rabbit muscle was examined by positive staining, negative staining, and freeze-etch electron microscopic techniques in the absence and presence of calcium transport conditions. The existence of 30-40 A particles covering the outer surface of FSR vesicles was confirmed by two different negative stains in unfixed, glutaraldehyde-fixed and osmium tetrox...
متن کاملProperties of the fragmented sarcoplasmic reticulum from fast twitch and slow twitch muscles.
Skeletal muscle of guinea pigs is composed of at least three types of fibers which are distinguishable by their kinetic and histochemical properties. Fast twitch muscles may be histochemically red or white; fragmented sarcoplasmic reticulum (FSR) prepared from both types of fast twitch muscles has identical calcium-accumulating capacity (maximal calcium uptake) and calcium-concentrating ability...
